Expression of apoptosis-related factors in psoriasis vulgaris

Psoriasis vulgaris is a type of chronic inflammatory keratosis characterized by epithelial hyperplasia. The aim of this study is to investigate the mechanism underlying this disordered keratinization with respect to cell death and cell proliferation in psoriasis. In psoriasis, Ki-67-immunopositive cells were observed in the suprabasal layers. Expressions of EGFR, Bcl-2 and Bcl-X (apoptosis-inhibiting factors) were strongly detected in all layers. The TUNEL assay demonstrated a considerable number of positive cells throughout all layers of the epidermis. However, we could not confirm apoptotic bodies via transmission electron microscopy (TEM). TNF-α and Bax (apoptosisinducing factors)-irnmunopositive cells were detected in all epidermal layers. Furthermore, dividing keratinocytes were observed in the suprabasal layers by TEM. Thus, we conclude that proliferation of keratinocytes predominates over cell death in psoriasis. Therefore, keratinocytes are considered to be stratified to the horny layer before keratinization.
