Great progress in decoding of human genome carries us into a post-genome century. A clear image can be drawn now concerning the apoptosis-related genes and their products, including the transduction of apoptotic signals. However, transcriptional control of the apoptosis-related genes still remains to be elucidated. One reason for this phenomenon is that multiple motifs for different transcription factors exist in promoter region of a gene. On the other hand, transcription factors can bind to their motifs in promoters of different genes. Therefore, transcription of a gene is controlled by complex mechanisms. We have focused on the human bcl-2 promoter, and here are our most recent results concerning transcriptional control of bcl-2 in normal human endometrium. As a preliminary study, the expressions and locations of the known transcription factors such as SP-1, SP-3, ATF-1, p53, and Brn3-a were observed by immunohistochemical staining of both the proliferative and secretory endometrium. Then, the binding activities of these molecules with their respective motifs in bcl-2 promoter were analyzed with the Biacore 2000 system. To accomplish this analysis, either the glandular or stromal nuclear extracts from the endometrium of different stages were injected over the senseor-chips coupled with Oligos containing different motifs. Transcription factor/motif binding was then revealed when rising curve was revealed following injection of a specific antibody. It is known that there is a cyclic expression of Bcl-2 in normal human endometrium. Corresponding to this expression pattern, we provided with evidence showing specific interaction between a member of AP-1, c-Jun and its motifs in bcl-2 promoter.
