In order to promote the bacterial growth and its oxidative activity, the initial pH of 1% yeast-extreact-medium was adjusted to 6.7 and after 3 day's pre-incubation at 24℃ the pH was reduced to 5.0 by the addition of substrate (mixture of Ca-l-idonate and Ca-d-gluconate, 5g per 100 c.c medium) and then the fermentation was carried out, adjusting the pH value to 5.0 every 24 hrs. In this case high yiels of 2-keto-hexonic acid was obtained, i.e., after 7 days it reached to 40.2% on the basis of the substrate, and mainly accumulated 2-keto-gluconic acid, while after 11 day's incubation main product was 2-keto-l-gulonic acid. Accordingly, the former was more easily formed and decomposed than the latter.In the second experiment, pre-incubation of the bacteria in d-glucose-yeast-extract medium containing inorganic nitrogen source was conoducted during 40 hrs. where vigorous respiration occurred and 2-keto-hexonic acid was hardly accumulated. Then the substrate was added after adjustment of pH to 5.0 and the successive fermentation was set going under the above described pH regulation.After 40 hrs, the production of 2-keto-hexonic acid reached to 51.5% on the basis of the substrate, and it comprised a larger amount of 2-keto-l-gulonic acid than 2-keto-d-gluconic acid.Pseudomonas fluorescences liquefaciens (Bacterial museum, KRAL), Pseudomonas fluorescens 33F (isolated by YOKOZAWA in 1950) and Pseudomonas fluorescens Y (isolated by the author in 1951) were compared in their activity producing 2-keto-hexonic acid and it was shown that the last strain was the most active, i.e., the yield was 50.3% 2-keto-hexonic acid on the basis of the substrate, after 3 days' incubation in the above described fermentation process.
