Recently the author has carried out a few further studies on the formation of 2-keto-hexonic acid from the mixture of Ca-l-idonate and Ca-d-gluconate by Pseudomonas fluorescens sp. in shake culture. The results may be summarized as follows : 1) Such low concentration of (NH_4)_2SO_4 as 0.1% is preferable for 2-keto-hexonic acid formation. High yield of that acid is attainable in the culture which has initiated at pH 6.1 and has been regulated in acidic side, pH 5.0,after a figorous multiplication of the cells.2) Oxalic and acetic acids were tested for the effect of buffer action. The yield of 2-keto-hexonic acid was 46.2% with the former and 47.8% with the latter. However, the acetic acid is favorable in facilities of observing the multiplication of bacterial cells and separating crystalline 2-keto-hexonic acid.3) When the concentration of inorganic salts is increased to six times, the formation of 2-keto-hexonic acid is lowered by 7〜10% than the usual. Thus the previously decided concentrations, i. e., 0.05g/100cc of KH_2PO_4 and 0.01g/100cc of MgSO_4. 7H_2O, may be preferable.4) The following N-sources are suited for the formation of 2-keto-hexonic acid as high as 6%, and the yields of crystalline Na-2-keto-l-gulonate are as follows.[table]
