The properties of alkaline proteinase obtained from Cephalosporium sp. ATCC 11550 were studied. The enzyme showed the maximam activity at pH 11 when incubated at 30℃. It also showed maximal activity at 50℃-pH 11 or at 60℃-pH 8.0. It was stable over the pH range 4 to 11 at 40℃ for 60 min. The enzyme powder showed nearly full activity at 45℃ after 17 weeks. The alkaline proteinase was markedly inactivated by di-isopropyl fluorophosphate (DFP), sodium lauryl sulfate (SLS), N-bromosuccinimide, potato inhibitor, Fe^<+++>, and Hg^<++>, whereas it was not affected by either metal chelating agents or sulfhydryl inhibitors. As this enzyme is a DFP-sensitive protease, it might be classified as a serine enzyme. Furthermore, it was elucidated that the enzyme possesses proteolytic specificities similar to those of chymotrypsin and trypsin, on the basis of investigations of the substrate specificity using various synthetic peptides.
