Basic research for the industrial production of thermostable protease was undertaken, and some strains of thermophilic spore-forming bacteria highly active in producing thermostable protease was isolated from soil. These strains grew aerobically and had an optimum temperature range for growth between 50 and 60℃.Employing a selected strain, crystalline thermostable protease was obtained from the culture filtrated by the procedure listed in the text.Some properties of the crystalline thermostable protease investigated were as follows.1) Tests by Tiselius electrophoresis method showed that the crystal consisted of single component.2) Elementary composition was as follows : C : 51.36%, H : 6.82%, N : 15.5%, O : 24.21%, P : 0.00%, Ash : 1.11%.3) The optimum pH of this enzyme was found between 7.0〜8.5; the stability pH range was pH 6.1〜9.0. 4) The protease was highly thermostable ; for example, about half of its original activity remained after heating at 80℃ for an hour in aquous solution, and practically no inactivation occurred at 50℃ or 65℃. Calcium ion protected this enzyme against heat inactivation.5) The protease was inhibited not by potato inhibitor but inhibited by EDTA.
