The kinetic and physiological implications of oxygen in growing bakers' yeast were investigated using aerobic and anaerobic chemostats. The following results werw obtained.1. The glycolytic enzymes investigated showed in all cases higher activity in anaerobic cells than in aerobic ones, which is understandable, because the main energy for anaerobic growth is obtained only by glycolysis.2. Substrate uptake rate was not affected by the presence of oxygen, though glycolysis rate was repressed.3. Analysis results of the enzymes led to the conclusion that the repression of glycolysis by oxygen in growing cells is a result of allosteric regulation at the phosphfructokinase (PFK) step. This supports Schatzmann's postulation that the Pasteur effect results from inhibition of PFK by ATP.4. No effect on substrate uptake during quick accumulation of glucose 6-phosphate (G6P) was observed in aerobic cells. It is therefore extremely doubtful that G6P induces a further feedback regulation.5. The fact that the repression of glycolysis by oxygen is weaker in growing cells than in resting ones should be explained by differences in quantities of effectors (NH_4^+, fructose 6-phosphate, etc.), because PFK is still allosteric sensitive in growing cells.
