Natural killer (NK) cells, the third major lymphocyte population, are important effecter cells against certain infections and tumors. They have also been implicated as a link between innate and adaptive immune responses. In recent years, much attention has been paid to the NK cell inhibitory receptors and their interaction with major histocompatibility complex class I molecules on target cells. On the other hand, traditional herbal medicine has been used to treat various patients in the field of primary care in Japan, and their immunomodulatory effects have been demonstrated. This review summarizes recent findings on regulation of NK cell activity with an emphasis on killer cell immunoglobulin-like receptors (KIRs) and the influence of traditional herbal medicine on the NK cell regulation.
