A simple and rapid method for analysis of Aconitum alkaloids in serum and urine using liquid chromatography electrospray ionization mass spectrometry

血清及び尿試料中のアコニチン系アルカロイド(アコニチン, メサコニチン, ヒパコニチン, ジェサコニチン)をExtrelut^[○!R]で抽出し, LC-ESI-MSで測定する迅速かつ簡便な分析方法を考案した。各アルカロイドの基準ピークイオンであるプロトン付加体をモニタリングイオンとして, カラムはXTerra^[○!R] RP18を使用し, 移動相は0.1% (v/v)ギ酸アセトニトリル溶液: 0.1% (v/v)ギ酸水溶液(24/76, v/v)の条件で各アルカロイドは良好に分離することができた。血清及び尿における直線性は絶対注入量として5-200 pgであり, 検出限界は絶対注入量として1 pg (S/N=3)であった。各アルカロイドの回収率は血清では83.7%〜89.0%, 尿では85.6%〜89.9%であり, また同時再現性及び日差再現性の結果は良好であった。本法はトリカブト中毒におけるアコニチン系アルカロイドの同定及び定量, さらにはヒトおけるアコニチン系アルカロイドの体内動態の検討に適応できるものと考えられた。加えて, 本法は迅速かつ簡便な方法である事から, 救急医療の現場においても十分有用な分析法であると考えられた。
