It is generally held that the rheumatoid factor (RF) is an antibody against aggregated human IgG and that the determinant group of the IgG consists in its heavy chain, Fc portion of its subunits. This paper was designed to investigate whether the reactivity of the RF is directed against the heavy chain, Fc portion of IgG in the native state or in the aggregated state. 1) According to the method of Fleischman, IgG was treated with 2-mercaptoethanol and the product was gel-filtrated through Sephadex G-200 in the N-acetic acid or phosphate buffer solution. IgG subunits obtained in this procedure were tested for their reactivities with RF. It was recognized that the heavy chain portion was strongly reactive with RF in its aggregated state even in the minimal amount. 2) Rabbit IgG was papain-digested according to Porter's method and was chromatographed by CM-cellulose. Among the fractions obtained, only the Fc aggregates were shown to be reactive with RF. These results seem to show that the determinant group of IgG against RF residues in the heavy chain, Fc portion in the aggregated state.
