Effects of Carragheenan on Radiation Lethality

The effect of carragheenan, a macrophage-toxic agent, on lethality of irradiated animals was studied in order to elucidate the possible participation of stromal elements on haemopoietic function. Treatment of mice with the agent resulted in an impaired differentiation of CFUc from CFUs which is considered to be responsible for the lower survival of the carragheenantreated animals after irradiation. Stromal cells are considered to play an important role in the control of the proliferation and differentiation of haemopoietic stem cells. Observations in genetically defective animals such as Sl/Sld mice have demonstrated that stromal elements are essential for the normal functioning of haemopoietic cells. Works by Maniatis et al. and by McCuskey et al. also showed the participation of stromal elements in the regeneration of haemopoietic system. We have reported that the 'blockade' of reticuloendothelial system (RES) with particulate substance enhances the survival and recovery of irradiated haemopoietic stem cells as well as the survival of the irradiated animals. Similarly, damages glven to RES would lead to an impaired recovery of the irradiated stem cells and the lower survival of the irradiated animals. In this regard, it would be of interest to investigate the effect on the radiation lethality of mice together with that on the survival of stem cells, since carragheenan has been reported to kill phagocyticcells specifically and eventually to suppress immune response.
