In Vitro Inhibition of Binding of Tumor Necrosis Factor(TNF)-α by Monoclonal Antibody to TNF Receptor on Glioma Cell and Monocyte

TNF-αによる有害な副作用はTNF-p75受容体(TNF-p75R)を介する反応が主体と考えられている。免疫染色により、培養グリオーマ細胞、グリオーマ組織および腫瘍周囲の脳組織において、TNF-p75Rは-p55Rに比べて発現が少ないことが示され、脳はTNF-αの局所投与に適した環境であると考えられた。抗TNF-p75R抗体はin vitroで単球系細胞U937上でTNF-αと競合的に結合し、U937からのTNF-αの産生を抑制するが、培養グリオーマ細胞GL-9においてはTNF-αによる抗腫瘍作用に影響を与えなかった。グリオーマに対するTNF療法において、p75受容体拮抗物質の併用投与によりTNF-αの有害作用が軽減できる可能性が示唆された。
