PRODUCTION OF STEROIDS IN ONE CASE OF GRANULOSA CELL TUMOR IN CULTURE

顆粒膜莢膜細胞腫は,ホルモン産生腫瘍として興味あるところであるが,産生細胞の局在および産生機序については未だ不明の点が多い.今回我々は,莢膜細胞の殆ど無い顆粒膜細胞腫を経験した.そこで前記の点を解明すべく,腫瘍より採取した組織片をトリプシン処理にてsingle cellとした後,TC-199,Hanks solution + 20%FCSの培養液中に単層培養を行ない,培養液中に分泌されたsteroid hormoneをRIA法により測定した.同時に細胞は,培養前後に,それぞれ,酵素組織学的,及び電顕的に観察した.培養中これらの細胞は,位相差顕微鏡下にて,全ての細胞が同一の形態をとり,上皮様パターンを示した.培養液中のestradiol-17βは術前患者血清中では,高値を示さなかったにもかかわらず,培養3日目に900pg/ml以上と非常に高い値を示した.dehydroepiandrosterone, progesterone及び170H-progesteroneは,比較的低値にとどまった.また酵素組織学的に,colony全体に3β-dehydrogenaseの存在を,電顕的には,steroid産生を示唆する所見が得られた.以上の成績より,顆粒膜細胞腫においては,顆粒膜莢膜細胞成分,あるいは間質細胞が,estrogen分泌の主体をなしているという説が以前より有力であったが,aromatase activityの存在も含め,顆粒膜細胞成分が,より重要な働きを有することが推察された.
