Light-Induced Denaturation of Bacteriorhodopsin Just above Melting Point of Two-Dimensional Crystal

The denaturation kinetics of bacteriorhodopsin indtrced by light irradiation was measured inthe temperattrre range between 80 and 86'C, which was just above the naelting point of the two-dimensional crystal cornposed of lcacteriorhodopsin 11101(2CLll(l6S in the ptrrple naeraabrane. Bacteri-orhodopsin was stable in the dark bvrt gradually denatvtred trnder the irradiation of visible lightlonger than 520 nm. Analyzing the Linne course of the light-indtrced denaturation phenoznenon,the kinetic constant of' the denaturation frorm a jchotointermediate state was estitnated. Thetemperature dependence of tlae kinetic constant lead to the efl'ective activatiora energy of abotrt411 kcal/tool. The stability of' a photointernaediate state was discussed on the basis of the light-induced denaturatiot'r behavior.
