EFFECT OF EXERCISE-TRAINING ON β-ADRENERGIC RECEPTORS OF RAT FAT CELLS

In the present study, (-)-(^3H)dihydroalprenolol ((^3H)DHA) binding data for fat cells were analyzed by the graphical fitting on the computer-generated figures based on the theoretical model of one ligand-two binding sites, and the effect of exercise-training (treadmill running) on β-adrenergic receptors of rat fat cells was investigated. The animals were divided into three groups : control, exercise-trained and pair-weighed. The exercise-trained rats ran on a rodent treadmill 5 days a week over 8 weeks. (^3H)DHA specific binding to intact fat cells was not saturated even at high concentration of (^3H)DHA. However, preincubating fat cells with 10μM valinomycin demonstrated saturable steady state of (^3H)DHA specific binding to fat cells. The Scatchard plots of these binding data displayed uniformly curvilinear with an upward concavity. These results suggest multiple populations of (^3H)DHA binding sites possessing different affinities. The calculated number of the high affinity binding sites per fat cells was similar in the three groups. The dissociation constant (K_D) of the high affinity binding sites for antagonist was also similar between control and the exercise-trained rats. In addition, the density of the high affinity binding sites expressed per cell surface area in the exercise-trained rats was about 2-fold higher than in control rats but not significant. It is concluded that exercise-training (treadmill running) can not affect the number of β-adrenergic receptors and the binding affinity for an antagonist in rat fat cells.
