65 IN VITRO CULTURE OF OROBANCHE RAMOSA, O. AEGYPTIACA AND O. MINOR

Massive seed germination occurred and Orobanche callii of various types, colours and growth rates were successfully induced, maintained and further enhanced. Compared to O. ramosa and O. aegyptiaca, O. minor was more difficult and took longer to produce callii. Better callus induction was obtained from B5 salts than MS media. The present protocol successfully produced callii with root-like protrusions, pad-like structures (with many papillae), possible terminal haustorium and secondary/adventitious haustoria, as well as haustorial hair. Drops of secretion were also observed at the tip of these protrusions. A new and simple aseptic "cover glass infection system"(CGIS) was developed and proved to be satisfactory.
