Presence of Acetone and Acetoacetate Decarboxylase in Soybean Root Nodules

Acetone was detected in the leaves, roots, and nodules of soybean [Glycine max (L.) Merr.]plants, but not in the stems. The concentration of acetone in the nodules was the highest among the three former organs. Although acetone was also detected in the leaves and the roots of non-nodulated soybean plants grown with nitrate, the concentration in each case was half of that in nodulated soybeans. A small amount of acetone evolved from detached nodules but not from nodulated roots. The activity of acetoacetate decarboxylase that catalyzes the conversion of acetoacetate to acetone was detected in the nodules, mainly in the bacteroids. These results suggest that acetone is produced by the action of acetoacetate decarboxylase in soybean nodules. The fluctuation of the acetone content in the nodules during their development were not correlated with those of the acetylene reduction activity. The correlation between the changes in the acetylene reduction activity and those of the acetone content in detached nodules was hardly apparent when detached nodules were incubated at different oxygen tensions. However, the level of acetone in the nodules was reduced when nitrate was supplied to nodulated soybean plants. The metabolism of acetone related to nitrogen fixation in soybean nodules was discussed.
