Abnormal Collagen Aggregates Induced in Rat Incisor Dentin after Sodium Fluoride Administration

Young male Wistar rats, weighing 100 g, were subcutaneously administered a single dose of 84 mg/kg of sodium fluoride. After 3, 6, 12, and 24 hours and 2 and 4 days, the rats were fixed by perfusion, and their upper incisors were examined by electron microscopy. In some instances, 0.1% ruthenium red was added to the fixative and demineralizing solution for the sake of observing the distribution of acid mucopolysaccharides. In the predentin, abnormal increases of amorphous ground substance occurred 3 hour after sodium fluoride administration and continued until 12 hours after administration. Thereafter, the increases gradually slowed; and the same level observed in eontrols resulted by the fourth day after administration. Ruthenium red staining showed this amorphous ground substance to contain large amounts of acid mucopolysaccharides. Two types of abnormal collagen, aggregate -segmental collagen aggregate (SCA) and fibrous long-spacing collagen (FLS) were observed in the predentin. In longitudinal sections, SCAs were about 300 nm long and 200-1000 nm wide; symmetrically arranged striations were visible within them. FLSs were observed as long, thick fibrils with periodic striations of about 150 nm. With the passing of time, these abnormal collagen aggregates shifted from the distal end of the odontoblast toward the dentin, in which they became embedded. Mineralization in the abnormal collagen aggregates commenced with deposition of needle-shaped or thin plate-like crystals. As time passed, the abnormal collagen aggregates came to be completely covered with the crystals. These findings suggest that abnormal collagen aggregates are produced as a result of the secretion from odontoblasts and are mineralized, as are normal collagen fibrils in dentin.
