Molecular Cloning of PCR Amplified BMP-Related Genes (GDF-5,6 and 7) from Rat Tooth Cells Using Modified pBluescript SK_+Vector

Growth differentiation factors (GDFs) were amplified from rat developing tooth organs and a rat dental pulp strain cell (RPC-C2A) by the PCR method. The amplified products were cloned into the modified pBluescript Sk_+ vector for TA cloning. This TA cloning system is prepared easily and is a useful rapid cloning method for PCR products. Nucleotide sequence analysis revealed that three types of GDF were cloned from the rat tooth organ and of a RPC-2A cell. These cDNAs were identified as GDF-5, -6 and -7 in comparison with the mouse GDFs. The results suggest that GDFS are highly expressed in the developing tooth organ and involved in tooth differentiation.
