4. Intracellular pH measurements in isolated rat osteoclasts.

The purpose of this study was to examine H~+ extrusion in rat isolated osteoclasts (OCs) by measuring intracellular pH(pHi) changes. pHi in OCs loaded with pH fluorescent indicator was measured on a confocal laser scanning microscope after acid loadiung. Those cells did not restore their pHi in Na^<A{>, K^<A{>-free meditnm, but pHi recovery was observed in Na^<A{> or K^<A{>-rich medium. These data indicate that OCs have mechanisms to recover pHi from an acid load to the resting level in a Na^<A{>or K^<A{>-dependent manner.
