A rapid detection method of pathogenic E.coli O-157 have been developed by using O-157 specific phage which formed clear plaque on them. Bacterium are first treated in the acidic buffer to kill acid-sensitive bacteria, then the surviving fractions of these bacterium including most of E.coli O-157 strain were amplified in the low pH(4.7)/citric-acid medium. Finally, O-157 specific phage mixture known to form clear plaque was spotted to comfirm the existence of E.coli O-157 in the starting materials. After the amplication of the test sample bacterium, clear spot is observed after five hours of incubation at 37℃. Because of these simple procedure,many sample can be tested with less expensive cost. This is very useful when we check numerous number of samples such as clinical sample, raw food materials and animal sample including beef etc.
