Pseudomonas aeruginosa secretes many kind of virulence factors, such as alginate, protease, siderophore, erastase, and biosurfactant (BS), and these secreted products cause to infect to human lungs. Recently, we have isolated and characterized new alginate regulatory gene, algH. Using algR2 and algH deficient mutant strains and complemented transformant strains, the expression levels of vilurence factors in P. aeruginosa were studied with various cultivation conditions. Big difference of alginate production in several strains were observed under liquid and solid medium. In the liquid cultivation, P. aeruginosa R2:: Cm carrying algR2 gene cultivated with PIB media secreted large quantities of alginate in the culture media, but not with LB media. The strain carrying algH gene produced the alginate with PIB, but the level was extreamly lower than that of carrying algR2. On the other hand, the P. aeruginosa carrying either algR2 or algH secreted alginate on solid media (PIA). These data indicate that the two regulatory genes are indispensable for high level of alginate production under liquid conditions, but either gene required for alginate production on solid media in P. aeruginosa. Protease, BS, and siderophore productions were also affected by both algR2 and algH genes. Especially the protease activities of strains carrying algR2 gene were very low levels.
