The effects of several staining reactions and treatments of the nucleus of the newt's erythrocyte are investigated, and the following results are obtained. 1. Each picture obtained by the three methods for the untreated sections, Feulgen's nuclear reaction, toluidinblue staining and thionin staining, coincides completely, but the sharpness of images obtained by the former two methods are much superior to the latter one. 2. The network structure observed in the picture by toluidinblue staining for the untreated sections, agrees well with the one stained by methylgreen in the methylgreenpyronin staining, 3. The network structure observed in the picture by toluidinblue staining for the untreated sections, does not necessarily coincide with the one stained by the hematoxylin. 4. The toluidinblue stainability is weakened when stained after the treatment in the 10% perchloric acid solution at 4℃ for 18 hours, compared with the untreated one, but no change is found in the structure itself. 5. The toluidinblue staining disappears perfectly after the treatment in the 10% perchloric acid solution at 70℃ for 20 minutes.
