The manganese-and copper and zinc-containing superoxide dismutase contents and their immunohistochemical localization in human thyroid tumors and some other thyroid diseases were examined and compared with those in the adjacent normal thyroid tissues. The enzyme-linked immunosorbent assay (ELISA) was used in this study for the measurement of Mn-SOD and Cu, Zn-SOD contents. The content of Mn-SOD tended to increase in diffuse hyperplasia and follicular adenoma. The content of Mn-SOD was significantly higher in papillary carcinoma than in the adjacent normal thyroid tissue. Follicular carcinoma also revealed a markedly high Mn-SOD content. The content of Cu, Zn-SOD tended to increase slightly in thyroid disease. The Mn-SOD and Cu, Zn-SOD activities were examined in thyroid diseases and also compared with those in the adjacent normal thyroid tissues. The SOD activity was measured by the xanthine/xanthine oxidase/nitrobluetetrazolium system in sodium carbonate buffer at pH 10.2. Mn-SOD and Cu, Zn-SOD activities were prone to increase slightly in thyroid diseases. However, no significant difference was found between the thyroid diseases and adjacent normal thyroid tissue in Mn-SOD or Cu, Zn-SOD activity. Follicular carcinoma and papillary carcinoma contained a significantly high Mn-SOD content compared with normal tissues. The Mn-SOD activity in these two carcinomas increased slightly. The discrepancy between the enzyme activity and immunoreacitive content suggested that an immunoreactive but enzymatically inactive or low active Mn-SOD protein existed in thyroid carcinomas. In the immunohistochemical study, the adjacent normal thyroid tissue showed granular positive staining of Mn-SOD in the cytoplasm, and also showed positive staining of Cu, Zn-SOD in the nucleus and the cytoplasm. To determine the proliferative ability in thyroid diseases, the stainings of proliferating cell nuclear antigen (PCNA) and epidermal growth factor receptor (EGFR) were examined. PCNA staining was evaluated by % labeling index (%LI) of positive staining cell nuclei. Increases in Mn-SOD, Cu, Zn-SOD, EGFR and %LI of PCNA were observed in the papillary proliferative lesion of diffuse hyperplasia. A strong positive stain of Mn-SOD was observed in papillary and follicular carcinomas, whereas in anaplastic carcinoma staining was markedly less intense in spite of heavy strong staining of EGFR and high labeling in %LI of PCNA. These findings indicate that the Mn-SOD content varies according to the degree of differentiation and generating ability of thyroid carcinoma. Therefore, the Mn-SOD content was studied in two vaiant lines of B16 melanoma ; B16F1 with a low metastatic potential and B16BL6 with a high metastatic potential. Using ELISA, the Mn-SOD content in the hyper metastatic B16BL6 cell line was significantly lower (P<0.01) than that in the B16F1 cells. This suggested that in the highly proliferative tumor for example an anaplastic thyroid carcinoma and malignant tumor in other organs, a disordering mechanism of Mn-SOD content elevation existed. An anaplastic thyroid carcinoma had a natural potential of proliferative ability, and the well differentiated thyroid carcinoma corresponded to normal thyroid tissue. Induction of Mn-SOD m-RNA was observed with TNF-α, IL-1β and IL-6. To investigate the elevation of Mn-SOD content in well differentiated thyroid carcinoma. The TNF-α, IL-1β and IL-6 content were studied in various thyroid diseases and thyroid carcinoma. There was no significant correlation between the Mn-SOD content and each parameter (TNF-α, IL-1β and IL-6 content) in each thyroid disease.
