To investigate the correlation between collagenase activity and invasion or metastasis of human thyroid cancers, type I and IV collagenase activities were measured using ^3H labeled collagens, and the immunohistochemical observation with anti-type IV collagen antibody and morphological measurement of the dimension of type I collagen dissolved by cultured cancer cells in the type I collagen gel were carried out. In papillary and follicular carcinomas with extracapsular invasion and lymph node metastasis, activities of type I and IV collagenase were higher than those in uninvolved or other diseased thyroid tissues. Moreover, these collagenase activities increased with the advance in the degree of TNM classification. In the immunohistochemical study, type IV collagen was exclusively localized in the basement membrane region of intact thyroid tissues. In thyroid carcinomas, type IV collagen disclosed the marked discontinuity or disappearance in the delineation of basement membranes. Moreover, immunostaining of the type IV collagen severely decreased in the surrounding intact tissue in thyroid carcinoma. In type I collagen gel embedded culture, cancer cells dissolved the surrounding collagens more than intact or other non-cancerous diseased cells. These findings suggested that increased activities of type I and IV collagenase played an important role in local invasion as well as metastasis in thyroid cancers, and also suggested that these collagenase were produced by thyroid cancers themselves.
