The effects of nitrous oxide on the lamina V type neuron of the spinal dorsal horn were studied in the rabbit. The animals were divided into two groups to examine the supraspinal influences ; an intact spinal state and a transected spinal state. In 75% nitrous oxide sroup, to investigate the interaction of nitrous oxide and opiate receptors, naloxone 1.0 mg/kg was administered intravenously. The extracellular recording from the lamina V type neuron of the spinal dorsal horn was carried out at the lumbar segment by using the tungsten microelectrode. The neuron was stimulated by the intra-arterial injection of bradykinin (1.0 μg/0.1 ml). Fifty percent nitrous oxide suppressed bradykinin induced activities in the intact state (43.0±9.2%, mean percent of control±SE, p<0.01 vs. control), but did not show no significant change in the transected spinal state. Seventy-five percent nitrous oxide suppressed the induced activities both in the intact spinal state (27.7±7.0%, p<0.01) and in the transected spinal state (52.1±4.7%, p<0.01). Spontaneous activities were suppressed in the intact spinal state, but not in the transected spinal state in 50% and 75% nitrous oxide groups. The suppressive effects of 75% nitrous oxide were partially reversed by naloxone. These results demonstrate that 50% nitrous oxide suppresses the bradykinin induced activity through its facilitatory action on the descending inhibitory system ; in 75% nitrous oxide the level of the spinal dorsal horn is suppressed directly. The suppressive action of nitrous oxide on the lamina V type neuron of the spinal dorsal horn is partially mediated the opiate receptor by releasing endogenous morphine like factors.
