Myocardial Contractile Force and Regional Coronary Perfusion with Hyperoxic Blood : A Paradoxical Effect of 100% Oxygen Inhalation

The mechanism of the decreased myocardial contractile force caused by oxygen inhalation was studied. The left anterior descending coronary artery was perfused with femoral arterial blood (pO_2 : 90±13mmHg) in 34 open-chest mongrel dogs, and then the artificial lung was set in the perfusion circuit for the perfusion with hyperoxic blood (pO_2 : 463±62mmHg). Myocardial contractile force was measured with a myocardial strain gauge arch sutured on the epicardial muscle of the perfused area. When the perfusing blood was changed from normoxic blood to hyperoxic blood, the myocardial contactile force was increased by 11.1±20.1% (p<0.001). When coronary perfusion rate was decreased by 18.9±4.7% provided the other parameters were the same, the increase of myocardial contractile force was reduced to 8.0±17.9% (p<0.05). If oxygen inhalation was given to the dogs in addition to the perfusion with hyperoxic blood in the regional myocardium, the myocardial contractile force was decreased by 4.l±7.7% (p<0.02). It is suggested that a decrease of myocardial contractile force is produced by a reduction of coronary blood flow and also by an inhibitory action elicited by hyperoxia of the whole body when oxygen was inhaled, possibly caused by an inhibition of sympathetic nerve activity.
