A monoclonal antibody (Ted-R-2) to the 55 kd, pI 4.7,acidic protein of the newborn rat epidermis was developed by immunizing mice with a pI 4.7,hematoxylin stainable fraction of the extract in a 50 mM Tris-HCl buffer, pH 7.3,containing 10μg/ml phenylmethylsulfonyl fluoride from the newborn rat whole epidermis. As the antigen was shown to be located on the keratohyalin granules in the indirect immunofluorescent study, this 55 kd, acidic protein is considered to be synthesized at the terminal differentiation of the keratinocyte. The major amino acids of the 55 kd protein were glycine (22.7%), serine (16.5%), glutamic acid (13.4%) and ornithine (6.2%), but the histidine residue was only 3.8%.
