A monoclonal antibody (Ted-R-1) was developed by immunizing mice with pI 4.7,a hematoxylin stainable fraction of the extract in a 50 mM Tris-HCl buffer, pH 7.3,containing 10μg/ml phenylmethylsulfonyl fluoride from the newborn rat whole epidermis. An indirect immunofluorescent technique revealed that Ted-R-1 was located in two places : 1) keratohyalin granules and 2) the cell membrane region of the stratum corneum. Further the immunoblotting analysis of pI 4.7,the antigenic fraction which was used to produce Ted-R-1,demonstrated that there was only a single 55 kd polypeptide with which Ted-R-1 could react. No positive spot was seen in the keratin proteins. The major amino acids of the 55 kd, pI 4.7 protein, were glycine (24.4%), serine (17.3%), glutamic acid (14.1%) and alanine (7.7%), but the histidine residue was only 3.1%. Judging from the amino acid composition, this protein is considered to be the third component of the keratohyalin granules.
