概要

デンドリティックポリリジンはアミノ酸から構成されるデンドリマーで,プラスミドDNAと中性の複合体を形成し,さまざまな培養細胞に対して,低毒性で高いトランスフェクション効率を示す.リニアタイプのポリリジンとくらべ,細胞内に取り込まれる量は少ないが,細胞質への移行効率が高く,また,DNAのコンパクションが弱い.細胞内でDNAをリリースしやすい,あるいは,転写因子がアクセスしやすいことが高発現の原因だろう.プラスミドDNAとの複合体を尾静脈より投与すると,3時間後においても分解されていないDNAが血中から観察され,その他,肝臓や肺にも分布がみられた.さらに,siRNAの肝臓へのデリバリーにも成功し,そのin vivoでの応用が期待される.
