The glycosphingolipid(GSL)which contained oligosaccharide as a sugar portion was extracted from Sphingomonas trueperi and purified by silica-gel column chromatography. The GSL contained D-galactose and D-glucosamine in addition to lipid and acidic sugar components. The lipid components were identified as 2-hydroxy-myristic acid and the two kinds of dihydrosphingosines by gas-liquid chromatography after derivatization. The molecular weight of the GSL was measured by MALDI-TOF mass spectrometry, and the anomeric configurations of the sugars were identified by 1H-NMR. The data of 1H-NMR were compared with those of other GSLs with oligosaccharides isolated from other Sphingomonas species and closely related bacteria, and the chemical structure of the GSL from S. trueperi was proposed. The structure of the sugar portion was found to be identical to that of GSL-3 isolated from Novosphingobium capsulatum.
