Cefozopran, a new cephem antibiotic: stability against and affinity for β-lactamases and antibacterial activity against β-lactamase producing bacteria

Cefozopran (CZOP) a novel semisynthetic cephalosporin, was highly resistant to hydrolysis by various β-lactamases except plasmid-mediated OXA-1 penicillinase and oxyiminocephalosporinases, and consequent-ly showed potent antibacterial activity against most β-lactamase producing bacteria as well as β-lactamase non-producers. CZOP had Km or Ki values greater than 500μM for most β-lactamases tested. Owing to the low affinity for and resistance to the β-lactamases, CZOP was also strongly active against <I>Enterobacter cloacae</I> and <I>Citrobacter freundii</I> strains which, by producing large amounts of β-lactamase, exhibited nonhydrolytic barrier-type resistance against β-lactams that had high affinity for β-lactamase.
