EFFICACY OF MEROPENEM AGAINST EXPERIMENTAL RESPIRATORY TRACT INFECTION AND INTRA-RENAL INFECTION IN GUINEA PIGS

The <I>in vivo</I> efficacies of meropenem (MEPM), a new parenteral carbapenem, against experimental respiratory tract infection and intra-renal infection in guinea pigs were compared with those of imipenem/cilastatin (IPM/CS) and ceftazidime (CAZ). MEPM was as effective as IPM/CS against pneumonia with 2 strains of <I>Pseudomonas aeruginosa</I> at a dose of 5 mg/kg, and was significantly more effective against one strain and equally effective against the other strain, compared with CAZ. In neutropenic guinea pigs, MEPM was as effective as IPM/CS and was significantly more effective than CAZ at a dose of 5mg/kg against pseudomonal pneumonia. MEPM also showed excellent efficacy against intra-renal infection with <I>P. aeruginosa, Escherichia coli, Proteus mirabilis</I> and <I>Citrobacter freundii</I> at doses of 1 or 5 mg/kg. Against P. aeruginosa, the activity of MEPM was significantly superior to that of IPM/CS and equal to or superior to that of CAZ. Against <I>E. coli</I>, all three drugs were equally effective. Against <I>P. mirabilis</I>, CAZ was the most effective, and MEPM was superior to IPM/CS. Against <I>C. freundii</I>, MEPM was the most effective, and IPM/CS was superior to CAZ.
