PHARMACOKINETICS AND CLINICAL EFFICACY OF CEFTRIAXONE (Ro 13-9904) IN SURGICAL FIELD

Pharmacokinetics and clinical studies were performed on ceftriaxone (CTRX, Ro 13-9904). CTRX was injected intravenously at a dose of 1 g to 5 patients. Blood samples were taken at 1, 3, 5, 6, 7 hours after dosage and bile from T-tube or PTCD tube at 1, 3, 5, 6, 7, 24 hours was collected. The assays were performed by a routine bioassay technique. The mean serum levels of CTRX at 1, 3, 5, 6, 7 hours after dosage were 92.2, 77.6, 58.9, 59.4, 44.7μg/ml, respectively. The mean bile levels of CTRX at 3, 5, 7 hours after dosage were 47.5, 43.5, 35.0μg/ml. Each sample showed higher concentration compared with those of cefmetazole (CMZ) by cross-over method. CTRX was administered intravenously at 1 g one time a day for 7 days to a patient with felon and an effective response was obtained.
