The bacteriological evaluation of T-1982, a new cephamycin antibiotic, was studied in comparison with that of cefmetazole and cefotetan. The following results were obtained.<BR>T-1982 showed a broad antibacterial spectrum against gram-positive and gram-negative bacteria as well as cefmetazole and cefotetan, and its antibacterial activity was almost the same as cefotetan in the sensitivity distribution to various clinical isolates.<BR>Moreover, T-1982 was active against <I>Bacteroidxs fragilis</I>, but was not active against <I>Pseudonumas aeruginosa</I>.<BR>T-1982 showed a dose-response bactericidal activity against <I>Escherichia coli, Klebsiella przeumoniae</I> and <I>Serratia marcescens</I>.<BR>The correlation between MICs and MLCs of T-1982 against <I>Escherichia coli</I> and <I>Serratia marcescens</I> was similar to that of cefmetazole and cefotetan.<BR>Therapeutic effect of T-1982 on experimental infection in mice with <I>Escherichia coli</I>, <I>Klebsiella pneumoniae</I> and <I>Serratia marcescens</I> was somewhat superior to that of cefotetan, but inferior to that of cefmetazole on infection with <I>Proteus morganii</I>.<BR>The morphological change in <I>Escherichia coil</I> K-12 was observed by a phase contrast microscope. At concentrations of 0.05 and 0.5μg/ml, T-1982 made the bacteria elongated.<BR>A study on the binding of T-1982 to the penicilin-binding proteins in <I>Escherichia coli</I> K-12 and <I>Pseudomonas aeruginosa</I> E-2 revealed that T-1982 had the highest affinity for PBP 3.
