Mezlocillin, a new semisynthetic penicillin, has a broad spectrum antimicrobial activity. The authors investigated serum levels, urinary excretion rate, clinical effectiveness and side effects of the antibiotic. The results obtained are summarized as follows:
Following an intravenous drip infusion of 2 g mezlocillin in a 200 ml of 5% glucose solution in 4 patients average peak level was 172.5μg/ml immediately after completion of intravenous infusion, decreasing rapidly thereafter within 4 hours. Urinary elimination of the agent was 988 mg (49.4%) within 4 hours.
Mezlocillin was given to 27 patients with superficial infections, abscesses, acute peritonitis, biliary tract and postoperative wound infections, and urinary tract infections by means of an intravenous drip infusion.
Response was excellent in 9 cases, good in 7, fair in 8 and poor in 3 (efficacy rate 88.9%).
Side effects seen in 4 cases were skin eruption in 1 case, transient elevation of serum GOT and GPT in 2 cases and thrombocytopenia in 1 case.
