EXPERIMENTAL AND CLINICAL STUDIES ON DL-8280 IN URINARY TRACT INFECTIONS

DL-8280, a new pyridone-carboxylic acid derivative was evaluated both experimentally and clinically on urinary tract infections, and following conclusions were obtained.<BR>1) In the 17 registered strains, species of which are usually found in urinary tract infections, spectra of DL-8280 were broad.<BR>2) The peak MICs of DL-8280 for ampicillin resistant <I>E. coli</I> (98 strains), and <I>P.aeruginosa</I> (51 (51 strains) and <I>S.marcescens</I> (28 strains) were fairly superior to that of NA.<BR>3) The peak sprum concentration after oral administration of 200mg of DL-8280 was 1.65μg/ml at 3 hours, and half life was 4.5 hours. The peak urinary concentration was 254μg/ml at 2-4 hours, and urinary concentration level of 32μg/ml was observed at 12-24 hours.<BR>4) Overall effectiveness rate of DL-8280 in acute simple cystitis was 100% and all cases showed excellent response.<BR>5) Overall effectiveness rate of DL-8280 in complicated UTI was 95%. Even in the groups with indwelling catheters and mixed infections, over all effectiveness rate of DL-8280 were 100% respectively.<BR>6) MICs of DL-8280 for strains isolated before treatments were less than 3.13μg/ml, and all strains were eradicated after treatments.<BR>7) No subjective side effects and abnormal changes of laboratory examinations were observed.<BR>In conclusions, DL-8280 was suggested to be effective and safe antibiotic for the treatments of UTI.
