The solubility of protected peptides as well as the coupling rate was quantitatively examined in highly polar solvents. Symmetrical anhydrides in dimethyl sulfoxide (DMSO) containing <I>N</I>,<I>N</I>-dimethylformamide (DMF) or DMF solution of lithium chloride and diethyl phosphorocyanidate with triethylamine in DMSO–DMF were proved to be efficient coupling reagents for solid phase synthesis of hardly soluble peptides.
