In the first paper of this series(1), we reported on the production of V. B12 by shaking culture of Propionibaeteriur freudenreich ATCC 6207 and recognized that the high yield of V. B12 could be obtained. As the first step of the large scale production, we used 30l-Jar-Fermentor and studied on the change of the composition of the medium during the fermentation together with the production of V. B12. The results obtained are shown in Table 1, 2, Figures 5 and 6, by which we find that it produces 2.4mg/l V. B12 on the culture for 96 hours. The medium used for V. B12 production was of the following composition: 300g glucose, 300g peptone, 300g dried yeast, 120g K2HPO4, 1 p. p. m. CoCl2.6H2O and 151 tap water. Then, 15cc soybean oil was added, adjusted to pH 6.8, autoclaved at 15 lbs. for 50 min., and inoculated with 5% seed culture and incubated on reciprocal shaker at 30° for 48 hours. During the fermentation, glucose converted into volatile acid, and the more the amount of volatile acid increased, the more V. B12 was produced. Favourable production of V. B12 is to be traced by the determination of volatile acid.
