These experiments were carried out, in succession to our previous researches, to decide whether or not eisenin has a diketopiperazine linkage, and it was concluded from the following results that eisenin is, as it was reported before, not a diketopiperazine compound but a peptide, l-pyrrolidonoyl-α-l-glutaminyl-l-alanine: 1. Nearly 1 mol equivalent of amino itrogen was liberated from l-pyrrolidonoylamide and l-pyrrolidonoyl-α-l-glutamyl-diamide by the partial hydrolysis with 3% Ba(OH)2 solution for 3 hours at 100° on a water bath. The same result as this had been obtained previously in the case of eisenin. But more excess amino nnitrogen, amounting to about 1.5 to 2 mol equivalents, was liberated from 2, 5-diketopiperazine and 2, 5-diketopiperazine-3, 6-diacetyl-diamide by the same hydrolysis. 2. l-Praline was recognized by chromatographical study on the substance which was obtained from eisenin by hydrolysis after reduction with ethyl alcohol and Na at boiling temperature, and the same fact was found also with l-pyrrolidonoyl-α-l-glutamyl-diamide by the same treatments as above.
