Delayed Implantation Induced by Anti-Estrogenic Compounds in Rats

Effects of anti-estrogenic compounds, ethamoxytriphetol [1-(<I>p</I>-2-diethylaminoethoxyphenyl)-1-phenyl-2 <I>p</I>-anisylethanol, MER-25] and epithioandrostanol (2a, 3a-epithio-5a-androstan-17β-ol, 10275-S), on ova implantation in postcoital female rats were investigated. Each of the compounds induced a delay in ova implantation when administered at the pre-implantation stage either in a single injection or in consecutive injections. The minimum effective dose required to induce delayed implantation was 1 mg a day for MER-25 and 0.25 mg a day for 10275-S, in 6 daily injections. The delayed implantation induced by these compounds was counteracted by a minutes amount of exogenous estrogen or pregnant mare's serum gonadotropin (PMS) given simultaneously with the anti-estrogen or at an appropriate time in the early stage of pregnancy. These results suggest that MER-25 antagonizes the action of endogenous estrogen in the uterus and thus induces delayed implantation. Since 10275-S has strong androgenic and antigonadotropic activities in addition to its anti-estrogenicity, it is assumed that, differing from the action of MER-25, 10275-S suppresses mainly the secretion, and subsequently the action, of estrogen by acting on all physiological steps at the hypo thalamic, pituitary, ovarian and uterine levels, thus inducing delay in ova implantation.
