The authors found that 0.2<I>N</I> sulfuric acid solution of galanthamine gave fluorescence by the treatment with potassium permanganate, and attempted to use the reaction for the fluorometric determination.<BR>The relative fluorescence intensity was affected by the concentration of sulfuric acid in the solution, the amount of potassium permanganate, temperature and time of the oxidation.<BR>From the extensive studies of the above conditions, a recommended procedure was given, in which 10 m<I>l</I> of 0.2<I>N</I> sulfuric acid solution of galanthamine (0.22.0 γ/m<I>l</I>) were treated for 2 hours at 5 ± 1°C with 0.01<I>N</I> potassium permanganate, decolorized with 2.0 m<I>l</I> of 0.01<I>N</I> oxalic acid, kept overnight, and the relative fluorescence intensity was measured.<BR>A linear relationship was found between the concentration and the relative fluorencence intensity, although the slant was a little different on each set of calibration line.<BR>Determination of galanthamine by this method should be carried out together with a simultaneous treatment of a standard sample as the reference to assure an accuracy.
