In the previous paper, it was shown that the greater number of protein digestive enzymes in the digestive tract originate from the posterior salivary gland. The present paper deals with isolation and purification of proteolytic enzymes from the gland.Results obtained are summarized as follows: a crude extract was obtained with 1% NaCl solution from the tissues and subjected to gel filtration of Sephadex G-75. From proteolytic enzyme fraction obtained by the gel filtration, seven proteolytic enzymes and carboxypeptidase A like enzyme (CPA like enzyme) were separated by ion-exchange chromatography on CM-Sephadex C-50. Three of the seven enzymes obtained were electrophoretically homogeneous and hydrolyzed casein strongly at near pH 7. On the other hand, CPA like enzyme, when further purified by the ion-exchange rechromatography, was also found to be electrophoretically homogeneous.
