Immunohistochemical localization of ethanol-inducible P450 II E1 in rat liver. Effect of dietary fat.

To assess the effect of dietary fat on the induction of P450 II E1 in ethanol fed rat livers, a liquid high fat diet (HFD) or low fat diet (LFD) with ethanol (EtOH) or isocaloric glucose was given orally to male Sprague-Dawely rats for 21 days. At sacrifice, P450 II E1, total P450, acetaldehyde and thiobarbituric acid (TBA) reactive substance levels of rat livers were measured. Immunohistochemical staining was performed using antibody against rat P450 II E1, and immunoreaction intensity was blindly rated in order to provide asemiquantitative assessment of P450 II E1 levels. In EtOH-HFD group, staining of P450 II E1 was found in perivenular, midzonal and periportal areas, though EtOHLFD group showed staining only in perivenular area. P450 II E1, total P450 and TBA reactive substance levels in EtOH-HFD group were significantly higher than respective values in other groups. These results indicate that the dietary fat may play an important role in the development and enhancement of alcoholic liver damage due to induction of P450 II E1.
