Mutagenic Activities of Bellidifolin, Methylbellidifolin, and Methylswertianin in the Methanol Extract from Swertiae Herba (Gentianaceae)

Three substances showing mutagenic activity in Salmonella typhimurium TA100 with S9 mix were isolated from methanol extract of Swertiae Herba (Gentianaceae) by conventional techniques, and were identified on the basis of analytical and spectral data as methylbellidifolin, methylswertianin, and bellidifoin. Their specific mutagenic activities of methylbellidifolin, methylswertianin, and bellidifolin expressed in terms of the number of the revertant colonies per pg were 17.8, 6.9, and 30.4, respectively. The mutagenic activity shown by Swertiae Herba may be considered mainly to be due to bellidifolin, because it is the major hydroxyxanthone (91% of the total hydroxyxanthones content) in Swertiae Herba and has higher specific mutagenic activity.
