Effects of Goreisan(五苓散)and Inchingoreisan(茵〓五苓散)on Alcohol Metabolism in Mice Liver

The active mechanisms of two Chinese traditional medical preparations, Goreisan (五苓散) and Inchingoreisan (茵〓五苓散), were investigated in relation to alcohol metabolism in the liver using mice fed on a high fat diet (35% fat as total calories) with 15 g/kg ethanol for 4 weeks. The lipid levels in the liver were then determined. The lipid peroxide (LPO), triglyceride (TG), phospholipid (PL) and total cholesterol (T-cho) levels were increased in the alcoholic liver fed on high fat diet with ethanol. Such alcohol-induced elevation of LPO and TG levels were shown to be significantly counteracted by the administration of Goreisan or Inchingoreisan. Alcohol-induced high PL level was lowered by the administration of Goreisan. Administration of Inchingoreisan had little effect on T-cho level. These results suggested that these preparations possess an anti-fatty liver effect. Administration of ethanol generally lowered the level of reduced glutathione (GSH) in liver and other organs, and the treatment with Goreisan or with Inchingoreisan appeared to counteract the effect produced by alcohol. Activities of the enzymes involved in glutathione metabolism (G-R, G-Px, G-6-PD, 6-PGD, G-ST, γ-GPT) and of alcohol dehydrogenase (ADH) and aldehyde dehydrogenase (A1DH) in liver were also determined. The two preparations were examined for their effect on such alcohol-induced affected enzyme activities. Goreisan and Inchingoreisan appeared to accerelate oxidation of ethanol through the activation of these enzymes.
