The author has undertaken studies on the curative effects of a combination of vitamin D, parotin and citric acid on experimental rickets in the rat. In order to evaluate the antirachitic effects of this combination, investigation was conducted on the bone citric acid levels as well as the bone salts metabolism of rats to which these agents had been administered singly or in different combinations. The findings are summarized as follows: (1) The rats in which rickets had been developed experimentally were grouped into 5 categories and each group was given different treatment : no treatment was given to group 1 which was used as control; vitamin D was administered to group 2; parotin was administered to group 3; vitamin D and parotin were administered to group 4; and vitamin D, parotin and citric acid were administered to group 5. In the amounts of the bone ash, calcium, phosphorus and magnesium of the distal end and shaft of the hemerus, group 5 manifested the highest levels. Lower levels were observed in groups 4, 2, 3 and 1 in that order, with no large difference existing between groups 3 and 1. This will serve to indicate that a combination of vitamin D, parotin and citric acid is the most effective of all the means under investigation. (2) The rats in which rickets had been developed were grouped into 5 categories, and different treatment was given to each group: no treatment was given to group 1 which was used as control; citric acid was administered to group 2; citric acid and parotin were administered to group3; vitamin D was administered to group 4; and vitamin D and parotin were administered to group 5. In the citric acid, bone ash, calcium, phosphorus and magnesium levels of the femur, the greatest increase was manifested by group 5 as compared to group 1, which was used as control. The increase in citric acid was markedly larger than that in inorganic substances. This might be an indication that the mechanism of ossification is working vigorously.The results of the experiments have led the author to conclude that it would be rational to adminster a combination of vitamin D, citric acid and parotin in the treatment of rickets, because they play an important role in facilitating the mechanism of ossification though their action mechanisms differ. Vitamin D facilitates the adsorption of calcium through the intestinal canal; absorption of phosphorus, especially of organic phosphorus, is facilitated by citric acid; and parotin is assumed to promote the transfer of calcium and phosphorus from the blood to bone.
