Anti-proliferative effects and induction of apoptosis by liquiritigenin in human hepatocellular carcinoma SMMC-7721 cells

Liquiritigeninは甘草から抽出されるフラボノイドの一種である。Liquiritigeninの抗腫瘍作用を検討し,考えられる機序を明らかにするために,様々な濃度のliquiritigeninで48時間SMMC-7721細胞を処理した。MTTアッセイを用いてSMMC-7721細胞に対するliquiritigeninの増殖抑制作用を調べた。Hoechst 33258染色により細胞の核クロマチンにおけるアポトーシスの形態変化が認められた。フローサイトメトリーによりアポトーシス率を測定した。ウェスタンブロット法を用いてp53,bcl-2およびプロカスパーゼ3を解析した。カスパーゼ3活性はカスパーゼ-3活性キットを用いて測定した。本試験において,liquiritigeninは用量および時間依存的にSMMC-7721細胞に対して増殖阻害作用が強いことが示された。また,iquiritigeninにより,アポトーシスのプログラム細胞死の典型的な特徴であるSMMC-7721細胞の縮小,膜のブレブ形成(blebbing),アポトーシス小体の形成が生じた。フローサイトメトリーの結果から,SMMC-7721細胞の48時間のliquiritigenin曝露後にアポトーシス率が上昇し,それぞれ1.19%,7.48%,7.40%,31.48%,41.17%であったことが明らかになった。ウェスタンブロット法の結果から,liquiritigeninの処理後に用量依存的にp53の発現レベルが増加することが示され,対照群と比べ,Iiquiritigenin(0.40mM)によりp53の発現が約9倍増大した。一方,liquiritigeninの処理後,bcl-2タンパクの発現は低下した。我々の結果から,liquiritigeninによりプロカスパーゼ3の発現レベルは有意に低下したが,カスパーゼ3の活性は増加した。最新の研究結果から,liquiritigeninは細胞増殖を抑制し,SMMC-7721細胞のアポトーシスを誘発することが示され,これはおそらくp53およびbcl-2の調節,その後の下流分子であるカスパーゼ3の活性化を介すると考えられる。
