It is well known that ciliates cease to proliferate above a certain population cell density in the stationary phase under the usual culture conditions. It has been also reported that, in the axenically cultured ciliate Tetrahymena, some liquid soluble substances released from the cultured ciliates have very important roles in cell proliferation, as does the pH in the culture medium. We have little information about the reduction factors of cell growth in phagotrophic ciliates. Our preliminary experiments suggested that the growth of the ciliate Paramecium, which is a representative phagotrophic ciliate, might not be affected by the pH of the culture medium. Thus, we examined whether or not the growth of the phagotrophic ciliate was affected by the food amount and/or population cell density. When the ciliate Paramecium was cultured with the lettuce infusion, including an initial food density of 9.5×10^5 to 7.3×10^7 bacteria/ml at 23℃, the ciliate population cell density reached about 950 to 1,590 cells/ml at the stationary phase. On the other hand, if the initial food density increased to 6×10^8 bacteria/ml, the population cell density reached about 3,900 to 6,800 cells/ml at the stationary phase. However, at the stationary phase the addition of 1.6×10^7 bacteria/ml as food material did not increase proliferation. On the contrary, when the cells derived from the stationary phase of the culture were washed and transferred to a fresh culture medium including 1.6×10^7 bacteria/ml as food material, they underwent additional cell division. These results suggest that the growth of the phagotrophic ciliates Paramecium might be affected not only by the amount of food but also by some factors released from the cultured ciliates.
