The morphogenesis of muscle spindles was electron microscopically investigated using the rat sternomastoid and sternothyroid muscles. These muscles of 291 foetuses of the Wistar Strain rats from 18 to 22 days' gestation were examined. At 18 days' gestation, two muscle spindles were first deteCted in the early morphogenetic forms. The one of two muscle spindles showed more primitive morphological characteristics. This consisted of one intrafusal muscle fiber(IF) and one myoblast being in contact with the former and an uni- or bilamellar capsule. Large terminal axons coming from an unmyelinated nerve fib er bundle were in contact with the IF. At 19 days' gestation, two IFs were present. Each of them had myofibrilles being definitely differentiated and was in contact with nerve endings at the equatorial region. At 20 days' gestation, the third IF was seen between the two large IFs, being in contact with both of them. The forth IF appeared at birth, having the same manner of appearance as the third. Sensory nerve endings became larger and more numerous in the course of development of spindles, but they were confined to the outer surface of IFs during the foetal life, i.e. they enclosed all the IFs as one bundle. At 19 days' gestation, sensory nerve endings ran straightly in parallel with the longitudinal axis of IFs. But at 22 days' gestation, they ran spirally enclosing the IFs as one bundle. A few sensory nerve endings, consisting of two or three terminal axons, were seen in the foetal spindles. Some cored vesicles, of 500-700A in diameter, and a small number of mitochondria were also encountered in both single and multi-axonal sensory nerve endings. The above mentioned multiaxonal sensory nerve endings and cored vesicles diminished in number, but the number of mitochondria increased in the course of development. The fusimotor innervation was not observed before birth. The capsule consisted at first of one or two layers and at birth of 4 to 8. The space between the IFs and the capsule became more extended in the course of development. The inner capsule enclosing the individual IF was not observed during the foetal life.
